Sample Requirements

The optimal quantity of starting material is listed below. If sufficient DNA/RNA/protein quantity and/or concentration is not available, please consult the Core personnel before submitting.

Illumina Sequencing:

Note: additional 3 to 4μL of samples are required for sample QC/QA

Genomic DNA Seq: 100 ng Genomic DNA (1μg to 3μg) in 50μl or less of water/TE buffer

Directional mRNA Seq: 1μg (500ng to 3μg) Total RNA in 50μl or less water/TE buffer

Directional Total RNA Seq Ribo Zero: 1μg Total RNA (0.1μg to 4μg) in 10μl or less water/TE buffer

Small RNA Seq: 1μg Total RNA (1μg to 5μg) in 5μl or less water/TE buffer

Low input mRNA Seq: 500pg Total RNA (500pg to 100ng) in 10μl or less water/TE buffer

ChIP Seq: ≥1ng ChIP DNA in 10μl or less water/TE buffer

Exome Seq: 1-3μg Genomic DNA in 50μl or less water/TE buffer (can be done at 200ng if necessary)

DNA Methylation sequencing: 1-3μg Genomic DNA in 50μl or less water/TE Buffer

Illumina Microarray:

Note: additional 3 to 4μL samples are needed for sample QC/QA

DNA Methylation Microarray: 1μL DNA in 40μl or less water/TE buffer

Genotyping Microarray: 200ng DNA in 4μl or less water/TE buffer

Affymetrix Microarray:

Note: additional 3 to 4 μL samples are needed for sample QC/QA

Gene ST Array, or Human Transcriptome Array: 150ng total RNA in 3μL RNase free water (minimum concentration 50ng/μL, see Affymetrix protocol here for details)

3' IVT Expression array: 300ng total RNA in 3μL RNase free water (minimum concentration 100ng/μL, see Affymetrix protocol here for details)

miRNA 2.0, 3.0, 4.0 array: 500ng total RNA in 8μL RNase free water (minimum concentration 62.5ng/μL) or enriched LMW RNA from ≥500ng of total RNA in 8μL RNase free water (see Affymetrix protocol here for details)

For other type arrays, please contact Microarray core for specific sample requirements.

SOMAscan Assay:

150µl (in two aliquots of 75µl) of Human EDTA plasma (preferred), Serum, or Citrate Plasma in 0.5ml Matrix 2D Barcoded Screw Top Tubes. Tubes can be picked up at the core facility or purchased from Fisher Scientific Cat # NC0685529. Biological samples higher than Biosafety Level 2 or radioactive samples, and samples with solids/precipitates are not accepted. Submitting clear and non-Hemolyzed samples are crucial to ensure the best outcome for your study.

Ion Torrent Sequencing:

Note: additional 3 to 4μL samples are needed for sample QC/QA

DNA Seq: 10 to 40ng of DNA in 5μl to 20μL water/TE buffer (minimum DNA concentration 2ng/μL)
